Abstract: Integrated cryo-fluorescence microscopy and focused ion beam milling (cryo-FM-FIB) is a state-of-the-art technique for capturing fluorescently labeled targets in biological specimens and retaining them in thin lamellae for subsequent cryogenic electron tomography. Though previous approaches have applied to visualize fluorescent targets in vitrified cells, a lack of precision in target localization precludes its broader application to routinely capture small and rare structures of interest in situ. Our current work focuses on utilizing a customized integrated tri-coincident imaging system that allows for simultaneous fluorescence imaging and milling to visualize challenging sub-diffraction limited fluorescent biological targets with high fidelity. We have successfully localized the microtubule organizing center (MTOC) in vitrified mammalian cells as a crucial organelle responsible for cell division and cell differentiation. Tomographic reconstructions also revealed organization of microtubules at the MTOC and interactions with other organelles.
